What
is HPTLC in chromatography?
HPTLC is an advanced, automated and sophisticated version of
TLC. HPTLC is also known as planar chromatography or flat bed chromatography.
So many modifications are dined in TLC.
Automated sample injection system, a Pressurised system for
mobile phase is the changes made into the HPTLC over the TLC.
Mobile
phase and stationary phase in HPTLC:
The plates with different support materials and sorbent layers
with different formats and thicknesses are used.
Plates with a sorbent thickness of 100-250μm are used for
qualitative and quantitative analysis.
Example of Stationary Phase: Glass, Polyester sheet, Aluminum
Sheet.
The mobile phase should be of high graded.
Chemical properties, analytes, and sorbent layer factors should
be considered while the selection of the mobile phase.
The use of a mobile phase containing more than three or four
components should normally be avoided as it is often difficult to get
reproducible ratios of different components.
