Resolution in Chromatography

Resolution means the separation of peak in a chromatogram.

In Chromatographic language the resolution is defined as quantitative measure of how well two elution peaks can be differentiated in a chromatographic separation.

The resolution depends on the selection of mobile phase, buffer composition, and flow rate of mobile phase, sample nature and volume.

Calculation It is defined as the difference in retention times between the two peaks, divided by the combined widths of the elution peaks.

Optimum resolution shall be achieved in specified time.

Resolution is affected by following parameters:

- Selectivity

- Efficiency

- Retention
